Different types of aggressive breast cancer identified | Research ...

Vanderbilt-Ingram Cancer Center researchers have identified six subtypes of an aggressive and difficult-to-treat form of breast cancer, called triple-negative breast cancer (TNBC).

In the July issue of the Journal of Clinical Investigation, Cancer Center Director Jennifer Pietenpol, and colleagues describe the molecular features of these six distinct subtypes and identify chemotherapies to which the different subtypes respond in cultured cells and animal tumor models. Knowing the specific subtype could help physicians determine which therapies would work best in patients with TNBC and also inform the discovery and development of new drugs to treat this form of breast cancer, the authors suggest.

TNBCs recur more often, have poorer prognosis

TNBCs account for 10 percent to 20 percent of all breast cancers and tend to be more aggressive than other types of breast cancer. ?It?s a pretty significant health problem from the standpoint that 11 percent of Caucasians, 17 percent of Hispanics, and 25 percent of African Americans have this type of breast cancer,? Pietenpol said.

While some patients with TNBC initially respond well to standard chemotherapy, these tumors are more likely to recur after treatment and have a poorer prognosis. Less than 30 percent of women with metastatic TNBC ? which has spread outside the breast ? survive five years.

The difficulty in treating these tumors stems from what they lack.

The term triple-negative breast cancer, explained Pietenpol, ?is just a definition of what (the cancer) isn?t.?

TNBC tumors lack the estrogen receptors (ER) and progesterone receptors (PR) that drive about 60 percent of breast cancers. They also show no amplification of another receptor, called HER2, which drives about 20 percent to 30 percent of breast cancers.

The absence of these receptors means that the tumors are unlikely to respond to hormone therapies like tamoxifen and to therapies targeted to HER2 like trastuzumab (Herceptin).

Research describes six distinct subtypes of TNBC

Postdoctoral fellows Brian Lehmann and Joshua Bauer along with biostatistician Xi (Steven) Chen identified 587 cases of TNBC among 21 publicly-available breast cancer data sets. They then analyzed the genomic data to uncover unique gene expression profiles ? sets of genes that are either turned ?up? or ?down? in the tumors.

Their analysis revealed six distinct subtypes ? two ?basal-like? types (BL1 and BL2) involving cell cycle and DNA damage response genes; two ?mesenchymal? types (M and MSL) driven by genes involved in cell differentiation and growth factor pathways; an ?immunomodulatory? (IM) group, driven by immune system genes; and a ?luminal? subgroup (LAR) driven by androgen (male sex hormone) signaling.

The researchers also identified cell lines representing each of these subtypes and tested various chemotherapies in development or in clinical investigation in the cell lines. They also implanted these cells into mice to generate animal models of these tumor subtypes.

They found that the BL1 and BL2 subtypes responded to cisplatin; the M and MSL subtypes responded to dasatinib and experimental drug NVP-BEZ235; and the LAR subtypes were sensitive to bicalutamide.

Discovery could lead to new treatments

The findings provide a way of distinguishing molecular differences in this diverse group of breast cancers and suggest molecular targets for each subtype that may inform drug discovery and development efforts.

?In our opinion, the big breakthrough is just being able to say ?this isn?t one disease,?? said Pietenpol.

Being able to distinguish distinct biological subtypes of TNBC could help guide the design of select clinical trials for subtypes of breast cancer, point toward new biomarkers for patient selection for a given therapy, and identify new targets for drug discovery, she said.

?This really is the first step in translating genomic information into personalizing therapy for women with a very difficult-to-treat breast cancer.?

Best Species of Fish for Stocking Ponds | Freshwater Fishing News

Across North America, landowners have a variety of choices when it comes to stocking private ponds with fish. Types of freshwater fish suitable for pond stocking include bass, sunfish, catfish, freshwater trout and many others.

When choosing which species of fish to stock in a pond, owners typically take into consideration their overall goals for the project. In many cases, fishing is a priority for landowners.

In all but the coldest regions, members of the bass and sunfish family are popular with pond owners. These include largemouth and smallmouth bass, rock bass, crappie, bluegill, shellcrackers, pumpkinseed, green sunfish or other species.

In addition to bass and sunfish, some land owners choose to stock other freshwater species such as rainbow trout, bullheads, channel catfish, pickerel, yellow perch, carp, suckers or others.

Each species can have impacts on the overall success of the pond. A few species, such as largemouth bass and bluegill are known to co-exist successfully. Others can fail when placed in the same body of water.

Typically, pond owners stock one or more top level predators, such as bass or pickerel, accompanied by a variety of forage species upon which larger fish can feed on.

Before stocking a private pond, most owners consult pond fish books for ideas on which fish species to include. State or provincial fish and game departments may also offer information concerning pond stocking. Many of these resources can save pond owners time, money and frustration.

In addition to stocking fish for sport or table fare, pond owners often choose to add a variety of plants and wildlife. A variety of aquatic plants are introduced into private ponds. Aquatic plants can provide shelter for fish and other pond life, food for waterfowl, or simply add to pond aesthetics.

In many areas, mosquitofish are essential additions to even the smallest of ponds. These tiny fish eat insect larvae in great quantities and can be extremely effective for mosquito control.

When aquatic plants become a nuisance, some pond owners add species of fish that are intended specifically to reduce plant life. Common carp and grass carp are both known for their ability to devour and reduce submerged aquatic plants. Although they are highly effective at plant reduction, these fish tend to stir up silt, making pond waters murky.

Crayfish are another possibility for pond stocking. Like carp, they devour aquatic vegetation, stirring up silt in the process. In some applications, they become forage for large fish and their tendency to hamper water clairty is kept in balance. Although they have a host of benefits, some species are not suitable for stocking in ponds where water clarity and fish production are prime goals.

Australia inquiry backs refugee disaster rescue (AFP)

SYDNEY (AFP) ? A government inquiry into a horrific shipwreck off Australia which killed some 48 refugees Wednesday praised the rescue operation, saying it was "extremely complicated" to detect approaching boats.

The Senate Committee found rescuers did everything they could to save lives after the rickety vessel, carrying an estimated 90 Iranian, Iraqi and Kurdish asylum seekers, smashed on rocks at remote Christmas Island in December.

Originating from Indonesia's Palau Panaitan island, the wooden boat was dashed against limestone cliffs in a wild storm at the far-flung Australian outpost, throwing people to their deaths, including women and children.

Though navy and border officials managed to pluck 41 survivors from the huge waves and another man scrambled to land, it was the worst refugee accident in Australian waters in a decade.

The committee said "the response to the horrific tragedy that took place on 15 December 2010 was professional, courageous and as effective as it could possibly be under the prevailing weather conditions".

"(Rescuers') courage in the face of very real personal risk certainly increased the number of survivors. Their actions were nothing short of heroic."

The inquiry's scope did not extend to whether authorities had known the boat was approaching before it was physically sighted from the island -- a key issue currently being examined in an inquest.

But the report said the boat, codenamed SIEV 221, had been "unexpected" and the weather -- "amongst the worst ever experienced on the island" -- rendered on-ship radar and electronic surveillance useless.

A land-based radar had been trialled following the shipwreck with little success, it added.

"The radar picture of the marine environment around Christmas Island is extremely complicated, as it results from a range of objects including waves, clouds and birds, as well as vessels," it said.

"Simply detecting an object with the radar is not sufficient to identify it as a small boat."

The committee also noted recent media reports that a man in immigration detention at Christmas Island who was in touch with some on board SIEV 221 had tried to alert authorities of its approach three hours before the crash.

"Given that these media reports have emerged at such a late stage, the committee did not take evidence on the issue during the course of its inquiry, and it notes that the veracity of the claims remains untested," it said.

The claims would be considered by the coroner, it said.

Sometimes people can worry that there are difficult times ahead. But I am here to launch a report that clearly demonstrates America is sitting on the greatest assets you could wish for. This report, entitled "Building a 21st Century Communications Economy," shows how America can usher in a second American Century.

Natural resources, such as oil, are becoming harder to access and more expensive to purchase.

Global oil demand is projected to grow, on average, more than 20 million barrels a day by 2030, driven mostly by emerging economies. For example, the increased consumption of India and China alone is equivalent to using the entire U.S. strategic oil reserves in just one month (according to the International Energy Agency).

Economic value will increasingly reside in bits and bytes, rather than in the atoms and molecules of products and commodities.

There is enormous potential for broadband communication to free millions of people from so much unnecessary commuting to meetings.

According to a 2008 report by Connected Nation, the U.S. could expect to gain $92 billion through 2.4 million jobs created or saved annually by investing in broadband.

Total Annual Economic Impact resulting in broadband investment for Missouri would be $2,501,367,723 and jobs created or saved annually of 48,592.

Environmental opportunity: The products and services of an advanced communications network - including teleconferencing, telecommuting and e-commerce - lower environmental impact by reducing our need to travel and by reducing the materials used to produce physical objects (e.g. e-books instead of books, video-streaming instead of DVDs and especially substitutes for newspapers).

One report (the Smart 2020 U.S addendum) showed that information and communications technology (ICT) could help the U.S. reduce emissions by an estimated 13-22 percent from business as usual projections and potentially achieve gross energy and fuel savings of $140 billion-$240 billion. Robust accounting of emissions is integral to achieving a successful 21st century communications economy to ensure that carbon emissions are accurately recorded and accounted for - a truly dematerialized economy should not be limited by geography and physical boundaries.

Leading local initiatives like the Climate Prosperity Project show how business can lead the way. And so can government. Yesterday I had the honor to be with Mayor Slay celebrating the leadership of the city of St Louis in establishing a baseline for carbon emissions, and selection by ICLEI (International Council for Local Environmental Initiatives) as one of 10 cities to help them develop common criteria by which all cities' sustainability efforts can be measured and evaluated.

Here in the heart of America, I hope we can, with this report, highlight again the Spirit of St Louis. That was the first airplane to link continents. At critical points in St. Louis history, you had four great means of commercial transport: River, rail, road and air.

Please think now about the all important fifth: Information and Communications Technology. This is the medium where America can lead, from AT&T to Apple, from Cisco to Google, from Microsoft to Facebook, eBay and Amazon. America already leads the communications century, with online teaching and entertainment assets producing huge export potential. Claiming Compensation for Professional Negligence

Any professional may be negligent when offering advice to, or performing services for clients, and where such negligence causes financial loss, the professional in question will be liable to pay compensation for the damage they have caused. When the services of a professional are engaged, the client has a right to expect they will operate with reasonable care and skill, to the standard one would expect from a competent professional in the same industry. To prove negligence it will be necessary to establish that a duty of care existed, and that the duty was breached in a negligent manner, directly causing real or anticipated financial loss. Where this has occurred the client may pursue a claim for damages, which will be paid out under the indemnity insurance policy that professionals must maintain.

Claims for negligence are most common against professionals whose poor advice or service can cause significant financial loss, including solicitors, accountants, financial advisers, property surveyors and architects. Both solicitors and accountants may breach the duty of care they owe clients in various different ways. Failure to act promptly on the part of a solicitor may result in a claim becoming time-barred, otherwise a claim may be dismissed automatically if deadlines for submitting documents are not met. Solicitors also regularly make costly mistakes when dealing with property matters, such as failing to renew a lease for example, or committing errors when transferring legal ownership (conveyancing). Is Fat Tails Insurance Worthless? | zero hedge

Regular Zero Hedge readers know about our fascination with fat tail insurance, which is particularly relevant in modern day central planning when nothing is as it should be and everything is as the central bankers determine it is, at least until such time as the general market calls their bluff and we see the kinds of six sigma dislocations that marked trading for months on end after the Lehman bankruptcy. Probably the best most recent example is our overview of the 5 black swans that keep Dylan Grice up at night, and the way to hedge against them (incidentally, these were Long-term deflation, a Chinese Hard Landing, Asset Bubbles, Hyperinflation, and of course, a Bond Market Blow-up). And while one should always be hedge against prevailing conventional wisdom, because more often than not the crowd is wrong, and with very disastrous consequences, GMO's James Montier, in a just released white paper asks the fundamental question (whose affirmative answer could put purveyors of fat tail insurance such Nassim Taleb's Universa fund out of business), whether fait tails insurance is even necessary if everyone is protecting for the very same selection of "black swans" (therefore confirming that the hedged against events are anything but a black swan). Specifically, in Montier's words: "Tail risk protection appears to be one of many investment fads du jour. All too often those seeking tail risk protection appear to be motivated by the fear of missing out (not fear at all, but greed). However, the surge of tail risk products may well not be the hoped-for panacea. Indeed, they may even contain the seeds of their own destruction (something we often encounter in finance ? witness portfolio insurance, etc). If the price of tail risk insurance is driven up too high, it simply won?t benefit its purchasers." His solution for the best tail hedge: cash. "In many situations, cash is a severely underappreciated tail risk hedge." And somewhat more philosophic: "When it comes to timing tail risk protection, a long-term value-based approach and an emphasis on absolute standards of value, coupled with a broad mandate (a wide opportunity set, or, investment flexibility, if you prefer) seems to offer the best hope." Too bad that one must also factor for career risk, which usually means that everyone does end up doing precisely the same trade no matter what, which ultimately brings everyone back to square one.

While Grice's paper on the nuances of actual hedging to various strategies is far more detailed, here is how one approaches this topic in Montier's world:

We are aware of three general groups of tail risk protection from which investors may choose.

1. Cash

This is perhaps the oldest, easiest, and most underrated source of tail risk protection. If one is worried about systemic illiquidity events or drawdown risks, then what better way to help than keeping some dry powder in the form of cash ? the most liquid of all assets. (There is much more on the joy of cash to come shortly.)

2. Options/contingent claims

Occasionally, the market provides opportunities to protect against tail risk as a by-product of its manic phases. A
prime example was the credit default swaps that were a result of the demand for collateralized debt obligations.
These instruments were priced on the assumption that there would be no nation-wide decline in house prices, and thus offered a great opportunity (even without the benefit of hindsight) for those who were concerned that such an outcome was more plausible than the market thought.

Here, one caveat stands out above all others: one must be cautious of the dangers of over-engineering in this area of tail risk protection. It is too easy to construct an option that pays out under a very specific set of circumstances, and to do so relatively cheaply. But, of course, such an instrument tautologically only pays off under the realization of those specific events, so the tighter the constraints imposed, the less use the option is likely to be as general tail risk protection.

3. Strategies that are negatively correlated with tail risk

For the specific type of tail risk (illiquidity events) under consideration here, long volatility strategies are often said to
be negatively correlated. The simplest example of such a strategy is just to buy volatility contracts (bearing in mind
the roll return will be negative given the upward-sloping term structure of volatility). In the recent crisis, a dollar-neutral long quality/short junk portfolio acted very much like a long volatility strategy (with the added benefit of an expected positive return, in contrast to most insurance options).

Why Montier is skeptical: in one phrase - tail risk herding, driving up the price of hedges.

Tail risk protection is not a magic bullet. The cost of tail risk insurance becomes very evident when compared to the more conservative equities plus cash strategy considered in the sample above. This highlights one of the most important lessons of the exercise: tail risk protection requires timing ? the ?when? of tail risk protection, if you will. In the sample above, running permanent tail risk insurance similar to that of Strategy 4 leads to what might be called deferred drawdowns, made clear in Exhibit 3.

Obviously there is much more to Montier's thinking which can be found in the full letter below, although we would make one caveat: in tail risk insurance, like in everything else, it only pays to put a trade on if one is early, and, needless to mention, if the trade is the proper hedge against the specific risk. If either of these two are off the table, the naturally the costs will be prohibitive. Alas, in this day and age when alpha is all too often ignored at the expense of levered beta pursuits, in which the investing community has realized the banker trick that it only pays to be wrong if everyone is wrong (and thus will need a government bailout if all fail at the same time), do not expect much original and independent thought. After all - in a regime of centrally planned Bernanke put-ism, the only thing that matters is economies of scale. Whether one is right or wrong, when there is no risk of failure, is completely irrelevant.

Dame Judi Dench has been awarded the Freedom of the City of London at a ceremony in the capital.

In a video interview with The Telegraph, the actress known to James Bond fans as 'M', said she was "thrilled" to have been given the accolade.

As the paper points out, "many of the traditional rights received with the award are now effectively symbolic, such as being allowed to drive sheep over London Bridge or being hanged with a silken rope."

The award, which is believed to date back to 1237, is still an "honour" for the Oscar winning actress.

She said: "I love London, it's not my city at all, York is my city, but I feel very honoured to be given this. I've been here since I was a student so I've had many, many years here and now I feel like I belong."

And Colton Harris-Moore, the Internet-famous Barefoot Bandit, pleaded guilty in Seattle last week. What's next for both of them?prison, namely?is relatively clear. But what was life like before they were caught? Here are six stories about fugitive living:

Three years after skipping town, Bulger was frustrating investigators and endearing himself to neighbors all over the country. He made a particularly good impression with Gautreaux family in Grand Isle, Louisiana, where he spent the winter in 1995 and 1996 with the girlfriend who led to his capture this week.

The Whitey Bulger who was branded a reputed killer, crime boss, and bank robber by the 1986 President's Commission on Organized Crime often shut off the Gautreaux television, lecturing them on how bad it was to expose children to violent shows, including the local news.

The family of murderer Malcolm Webster's first wife have asked him to hand over her grave.

Claire Morris' brother Peter wants to replace her headstone in Aberdeenshire, which still bears her married name and refers to a "dear wife".

Webster was found guilty last month of murdering the 32-year-old in a staged car crash in 1994.

Mr Morris asked Webster, through his solicitor, to hand over the lair - but the request was rejected.

Aberdeenshire Council had earlier confirmed that changes to the gravestone, at Tarves, needed the authority of the owner.

Mr Morris initially said there was no way he would ask Webster, of Guildford, Surrey, to hand over the lair and appealed to the council to step in.

But now, with the backing of several politicians, Mr Morris plans to use human rights laws to force the killer's hand.

He said he had made the request to Webster so that all avenues had been explored first.

If the human rights option is unsuccessful, Mr Morris wants to exhume his sister's body and take it back to their family home in Kent.

He told BBC Scotland: "I swallowed my pride and my dignity, so I did make contact with Malcolm Webster's solicitor, who informed me that Malcolm Webster still claims that he is innocent and that he does not wish to give up Claire's grave."

Webster was found guilty of murder after a long-running trial at the High Court in Glasgow.

He was also convicted of trying to murder his second wife in New Zealand in 1999.

He was also found guilty of intending to bigamously marry another woman, Simone Banarjee, from Oban, Argyll, to gain access to her estate, while pretending he had leukaemia.

The 52-year-old, who denied the charges, is due to be sentenced on 5 July.

Police are also investigating concerns over the deaths of three children at a hospital in Abu Dhabi where Webster worked in the 1980s.
